Antibody Immunogen Description: Corresponds to amino acid residues N933 to L939; in the C-terminal of the kinase after the catalytic domain. Y936 phosphorylation stimulates phosphotransferase activity and induces interaction with APS, Cbl, Grb2 and Grb7. Phosphorylation also induces receptor internalization and protein degradation. This is the major in vivo phosphorylation sites in Kit (≥107 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus). Kit is known to be phosphorylated at this site in vitro by Kit.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.This antibody was also subject to negative purification over phosphotyrosine-agarose.

Scientific Background: Kit (SCFR) is a protein-tyrosine kinase of the TK group and PDGFR family, and regulates cell growth, survival, and differentiation. It is a receptor kinase that binds SCF (stem cell factor)/KitLG. It features an extracellular ligand-binding domain (five immunoglobulin-like domains), a transmembrane segment, a juxtamembrane domain, and an intracellular kinase domain. Mast, stem cell growth factor (steel factor) binding induces Kit dimerization and autophosphorylation. Protein interactions are induced with phosphorylation at Tyr-568 (with APS, Cbl, SHP2, & Src), Tyr-900 (with Crk & PIK3C2A) and Tyr-936 (with APS, Cbl, Grb2 & Grb7). Phosphorylation of Ser-741 and Ser-746 inhibits the kinase activity of Kit. Kit can mediate cell proliferation, survival, hematopoiesis, gametogenesis, stem cell maintenance, mast cell development, function, and migration, as well as melanogenesis. Some substrates that Kit can phosphorylate include PIK3R1, PLCG1, SH2B2/APS and CBL.This kinase is highly expressed and widely distributed in most tested human tissues. KIT is especially found in hematopoietic stem cells, mast cells, melanocytes, germ cells, and interstitial cells of Cajal. It was first identified as the cellular homologue of the feline sarcoma viral oncogene v-kit. Gain of function mutations in Kit are associated with gastrointestinal stromal tumours (GIST), mast cell leukemia, acute myelogenous leukemia (AML), and piebaldism.
