Antibody Immunogen Description: Corresponds to amino acid residues K171 to G177; in the protein kinase catalytic domain activation T loop region between subdomains VII and VIII. S174 phosphorylation stimulates phosphotransferase activity. This is one of the minor in vivo phosphorylation sites in KHS1 (≥6 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

It features a conserved N-terminal kinase domain and a large C-terminal regulatory domain, with a molecular mass of approximately 135 kDa. KHS1 interacts with adapter proteins like Crk and CrkL. A mutation in KHS1, K49R, inhibits the kinase phosphotransferase activity and inhibits downstream JNK proteins. It is involved in TNF-alpha-induced signaling in lymphocytes, such as Jurkat T cells. KHS1 links the Bcr-Abl oncogene and Ras to the stress-activated protein kinase pathway. It is ubiquitously expressed, with higher levels reported in the testis, ovary, and prostate. It is primarily found in the cytoplasm, with some localization to the plasma membrane and primary. KHS1 has been linked to diseases like Spastic Paraplegia 3A and Septal Myocardial Infarction.
