Antibody Immunogen Description: Corresponds to amino acid residues P593 to P599; in the EphA2_TM region between the transmembrane and kinase catalytic domains. Y596 phosphorylation stimulates phosphotransferase activity. This is one of the major in vivo phosphorylation sites for EphB4 (≥223 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

It features an N-terminal extracellular ligand-binding domain (composed of a globular domain, a cysteine-rich region, and two fibronectin type III repeats), a single transmembrane segment, and a cytoplasmic tyrosine kinase domain containing a SAM domain and a PDZ-binding motif. It binds the ephrin-B (EPNB) family of ligands (ephrin-A1, B2, or B3) on adjacent cells to mediate cell-contact signalling. It plays a critical role in vascular development, and angiogenesis. It acts as a cell surface receptor for mediating bidirectional signaling, controlling cell migration, adhesion, and separation. It is crucial for heart morphogenesis and arteriovenous differentiation during blood vessel development. EphB4 forward signaling typically promotes cellular repulsion and organization, separating EphB4-expressing cells from ephrin-B2-expressing cells. EFNB2 binding to EphB4 can be inhibited with a L95R mutation. High expression of EphB4 is often associated with tumorigenesis, particularly in colorectal, gastric, and prostate cancers, where it promotes proliferation and angiogenesis.
