Antibody Immunogen Description: Corresponds to amino acid residues E334 to S341; in the protein kinase catalytic domain activation T loop region between subdomains VII and VIII. S337 and T338 phosphorylation are predicted to be stimulatory for phosphotransferase activity. The phosphorylation of this S337 in CLK1 has not been reported in high throughput mass spectrometry studies, whereas phosphorylation of the T338 has been described at least twice.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.This antibody was also subject to negative purification over phosphotyrosine-agarose.

Scientific Background: CLK1 is a protein-serine/threonine kinase of the CMGC group and CLK family, and the LAMMER family, which is characterized by a specific EHLAMMERILG motif. It is a dual-specificity kinase that phosphorylates serine- and arginine-rich (SR) proteins (e.g., SRSF1, SRSF3) of the spliceosomal complex and may involve in regulatory mechanisms of RNA splicing. It controls alternative splicing, influencing gene expression, cellular stress responses, and viral replication. The phosphorylated serine/arginine-rich (SR) proteins involves in the pre-mRNA processing and releasing through the nucleus into nucleoplasm. It is primarily found in the nucleus, particularly in nuclear speckles, and recruited to nuclear stress bodies during stress. It contains a central kinase domain (residues 161-355) and an N-terminal region responsible for nuclear speckle localization. CLK1, which could phosphorylates specific SR proteins such as ASF/SF2, may directly regulates the activity and compartmentalization of SR splicing factors. CLK1 interacts with PPIG. This kinase shows high variability in human tissue distribution with the highest levels detected in adrenals, brain, kidney, liver, lung, ovary, pituitary, prostate, spinal cord, testes, thymus, thyroid, trachea, and uterus.
