Antibody Immunogen Description: Corresponds to amino acid residues E21 to V27; in the protein kinase catalytic domain in subdomain I.Y24 phosphorylation is inhibitory for phosphotransferase activity. This phosphosite is located in the kinase subdomain I region, which is involved in binding the phosphates in the ATP substrate. This is one of the major in vivo phosphorylation sites in CDK6 (≥243 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus).

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

It is involved in cell cycle regulation and differentiation, which promotes G1/S transition. It regulates cell proliferation, differentiation, and tumorigenesis by phosphorylating the retinoblastoma protein (Rb). Beyond cell cycle control, CDK6 plays a role in differentiation, prevents senescence, and aids in hematopoiesis. Its activity is controlled by association with D-type cyclins (D1, D2, D3) and inhibition by the INK4 family of inhibitors (e.g., p16). Phosphorylation at Thr-177 increases phosphotransferase activity. It is primarily active in the nucleus of proliferating cells, although it isfound in the cytoplasm of some cell types. Germline mutations in CDK6 are hypothesized to lead to a predisposition to melanoma. A197T mutation is found in family with autosomal recessive primary microcephaly-12 (MCPH12).
