Antibody Immunogen Description: Corresponds to amino acid residues V177 to G183; in the protein kinase catalytic domain. T180 phosphorylation stimulates phosphotransferase activity. This phosphosite is located in the kinase activation loop between catalytic subdomains VII and VIII. This is the major in vivo phosphorylation site in CaMK1d (≥19 reports from high throughput mass spectrometry studies recorded in PhosphoSitePlus). CaMK1d is known to be phosphorylated at this site in vitro by CaMKK1.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: CaMK1d is a protein-serine/threonine kinase of the CaMK group and CaMK1 family. It is activated by Ca2+/calmodulin. The binding of calmodulin is thought to result in a conformational change and can lead to subsequent activation through phosphorylation by CaMKK1 and CaMKK2. At least in part, it can also be activated by CaMKK1 in a calcium-independent manner. It is associated with calcium-triggered CaMKK-CaMK1 signalling cascade and activates CREB- and CREM-BETA-dependent gene transcription. It is highly expressed in polymorphonuclear leukocytes (PMNs). It regulates neutrophil function (granule function, chemotaxis, and respiratory bursts), promotes basal dendritic growth of hippocampal neurons, and is associated with insulin secretion in pancreatic beta-cells and linked to type 2 diabetes risk. CaMK1d also plays a role in the apoptosis of erythroleukemia cells and it has been linked to immune resistance in tumour cells.
