Antibody Immunogen Description: Corresponds to amino acid residues L186 to G192; in the protein kinase catalytic domain activation T loop region between subdomains VII and VIII. T205 phosphorylation stimulates phosphotransferase activity. This phosphosite is located in the kinase activation loop between catalytic subdomains VII and VIII. This is a minor in vivo phosphorylation site in BRSK1 (≥1 report by mass spectrometry). BRSK1 is known to be phosphorylated at this site in vitro by BRSK1, CaMK2a, and LKB1.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: BRSK1 ((Brain-Selective Kinase 1) is a protein-serine/threonine kinase of the CAMK group and CAMKL family. It is primarily expressed in the brain, where it plays a critical role in establishing neuronal polarity, axon branching, and synaptic development, as well as testis. It is located in the cytoplasm, nucleoplasm, cell junctions, and synaptic vesicles. It acts downstream of LKB1 (STK11), playing key roles in cell cycle checkpoint signaling (G2/M) and centrosome duplication. It is activated by phosphorylation on Thr-205 by LKB1 in complex with STE20-related adapter-alpha (STRAD alpha) pseudo kinase and CAB39. This can be prevented by an inhibitory T189A mutation. Its substrates include CDC25B, CDC25C, MAPT/TAU, RIMS1, TUBG1, TUBG2 and WEE1. Its phosphotransferase activity is lost with the K59A, and G327A mutations. It is involved in the UV-induced DNA damage checkpoint response, likely by inhibiting CDK1 activity through phosphorylation and activation of WEE1, and inhibition of CDC25B and CDC25C.
