Scientific Background: Bmx (Etk) is a protein-tyrosine kinase of the TK group and Tec family. It is a non-receptor kinase involved in actin reorganization, and in cell proliferation, survival, apoptosis, and cell adhesion in response to growth factors and interleukins. Another key functional role of Bmx is in TNF-alpha induced angiogenesis. Phosphorylation at Y566 by autocatalysis or by the protein-tyrosine kinase Src leads to activation and is required for STAT3 phosphorylation by Bmx. Src-induced Bmx phosphorylation could be almost completely inhibited with a Y566F mutation. Bmx has been demonstrated to prevent apoptosis of nasopharyngeal carcinomas.
