Antibody Immunogen Description: Corresponds to amino acid residues L435 to R441; in the region between the kinase catalytic and Ank_2 domains. The effect of S438 phosphorylation is unclear. It is the major site of in vivo phosphorylation in ANKRD3 with ≥12 reports by mass spectrometry in PhosphoSitePlus.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: ANKRD3 (RIPK4) is a 86-112 kDa protein-serine/threonine kinase of the TKL group and RIPK family. It is involved in keratinocyte differentiation, skin development, and wound repair. It is highly expressed in skin and hair follicles. It features an N-terminal kinase domain, a central region with 10 ankyrin-like repeats, and a C-terminal domain, and is often found at membranes. ANKRD3 interacts with Protein Kinase C (PKC). Phosphorylation at Thr-184 increases the phosphotransferase activity. Insertional mutagenesis studies in mice support a role for this protein kinase in mouse cancer oncogenesis. Mutations or dysfunction are associated with skin disorders, and it has context-dependent roles as both a potential oncogene or tumour suppressor.
