Antibody Immunogen Description: Corresponds to amino acid residues T312 to P318. Y315 phosphorylation is predicted to be inhibitory for phosphotransferase activity, although it has been suggested to be activatory. This phosphosite is located in the kinase activation loop just before the catalytic subdomain VIII. There has been ≥15 reports of its phosphorylation in high throughput mass spectrometry in PhosphoSitePlus. Akt1 is known to be phosphorylated at this site in vitro by Brk, Ret, and Src.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.This antibody was also subject to negative purification over phosphotyrosine-agarose.

Scientific Background: Akt1 (PKBA) is a protein-serine/threonine kinase of the AGC group and Akt family. It is normally inactive in the cytoplasm. When PI3K is activated by extracellular growth factors, Akt1 is recruited to the cell membrane and phosphorylated by PDK1 (T308) , mTOR (at S473) and other protein kinases to become active. Phosphorylation of T72, S124, S129, S246, T308, Y315,Y326, T450, S473 and Y474 all contribute to increased Akt1 phosphotransferase activity. TRAF6-induced 'K63'-linked Akt1 ubiquitination is critical for phosphorylation and activation. Phosphorylation of T34 inhibits phosphotransferase activity.
