Antibody Immunogen Description: Corresponds to amino acid residues D306 to V312; in the end of the Cofilin_ADF domain. The major site of phosphorylation of A6. The effect of Y309 phosphorylation is unclear. It is a major site of phosphorylation of A6 in mass spectrometry studies (>146 reports).
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: The A6 family of atypical kinases, which includes human A6 and A6r genes and their homologs, are often classified as tentative pseudogenes or putative protein kinases, with evidence for actual kinase activity being limited. Human A6 (Twinfilin-1) was initially reported to have tyrosine phosphotransferase activity. However, the closely related A6r (Twinfilin-2) was shown to bind ATP but failed to demonstrate kinase activity, indicating that some members may be functionally distinct or inactive (pseudogenes). These proteins have evolutionary homologs in flies, worms, and yeast. A6 also appears to be an actin-binding protein involved in motile and morphological processes. It inhibits actin polymerization, probably by sequestering G-actin. By capping the barbed ends of filaments, it also regulates motility.
