Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: PHLPP2 (PH domain leucine-rich repeat-containing protein phosphatase 2) is proposed to be a protein phosphatase involved in regulation of Akt and PKC signaling. It mediates dephosphorylation in the C-terminal domain hydrophobic motif of members of the AGC Ser/Thr protein kinase family; it specifically acts on 'Ser-473' of AKT1, 'Ser-660' of PRKCB isoform beta-II and 'Ser-657' of PRKCA. Akt regulates the balance between cell survival and apoptosis through a cascade that primarily alters the function of transcription factors that regulate pro- and anti-apoptotic genes. Dephosphorylation of 'Ser-473' of Akt triggers apoptosis and decreases cell proliferation. It also controls the phosphorylation of AKT3. It dephosphorylates STK4 on 'Thr-387' leading to STK4 activation and apoptosis. It dephosphorylates RPS6KB1 and is involved in regulation of cap-dependent translation. It inhibits cancer cell proliferation and may act as a tumour suppressor. Dephosphorylation of PRKCA and PRKCB leads to their destabilization and degradation. It dephosphorylates RAF1 inhibiting its kinase activity. Purified PHLPP2 does not appear to have Akt phosphatase activity, and it is proposed that it might be a pseudo-phosphatase. It is a zinc-binding protein, though it has been characterized as a phosphatase with unique structural characteristics compared to other PPM family members. It features a Ras association (RA) domain, a Pleckstrin homology (PH) domain, leucine-rich repeats (LRR), a PP2C phosphatase domain, and a C-terminal PDZ-binding motif.
