Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: DUSP12 (Dual Specificity Phosphatase 12; hYVH1) is an atypical, evolutionary conserved phosphatase that acts as a survival factor protecting cells from stress and apoptosis. It is related to the DUSP family of dual specificity phosphatases that target both serine, threonine and tyrosine phosphorylated sites in proteins. It is also known as a Glucokinase-associated dual specificity phosphatase (GKAP), and glucokinase appears to be a substrate. DUSP12 contains a catalytic dual-specificity phosphatase domain and a unique C-terminal C2H2 zinc-finger domain, which is crucial for its function. It lacks the N-terminal CH2 domain found in typical Mitogen-Activated Protein Kinase Phosphatases (MKPs). As a Saccharomyces cerevisiae YVH1 orthologue, it regulates ribosome biogenesis, cell cycle progression (G2/M). DUSP12 can inactivate stress-activated MAP kinases such as JNK and p38, suppressing proinflammatory cytokines. It interacts with ZNF622, a protein involved in regulating cell cycle, and is implicated in ribosome maturation. Found primarily in the nucleus, it is ubiquitously expressed, with highest levels in the spleen, testis, ovary, and leukocytes. It promotes cell proliferation and motility, with high expression reported in various malignancies. It also has potential links to type 2 diabetes susceptibility.
