Product Type Specific: SHIP1 (INPP5D) phospholipid phosphatase pan-specific antibody

Antibody Code: NP044-2

Antibody Target Type: Pan-specific

Protein UniProt: Q92835

Protein SigNET: SHIP1

Antibody Type: Polyclonal

Antibody Host Species: Rabbit

Antibody Immunogen Source: Human SHIP1 (INPP5D) sequence peptide Cat. No.: PE-01BBQ70

Antibody Immunogen Sequence: CKELYGEVIRTLPSLE

Antibody Immunogen Description: Corresponds to amino acid residues C217 to E232.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Antibody Modification: Unconjugated. Contact KInexus if you are interest in having the antibody biotinylated or coupled with fluorescent dyes.

Antibody Concentration: 1 mg/ml

Storage Buffer: Phosphate buffered saline pH 7.4, 0.05% Thimerasol

Storage Conditions: For long term storage, keep frozen at -40°C or lower. Stock solution can be kept at +4°C for more than 3 months. Avoid repeated freeze-thaw cycles.

Product Use: Western blotting | Antibody microarray

Antibody Dilution Recommended: 1 µg/ml for immunoblotting

Antibody Species Reactivity: This antibody detects the target phosphoprotein in the following species due to conservation of amino acid sequence: Human | Rat | Mouse.

Antibody Positive Control: The observed molecular mass of the processed target protein on SDS-PAGE gels is reported to be around 132-145 kDa.

Antibody Specificity: Very high

Scientific Background: SHIP1 (SH2 domain-containing inositol 5-phosphatase 1; INPP5D) is a phosphatidylinositol (PtdIns) phosphatase that specifically hydrolyzes the 5-phosphate of phosphatidylinositol-3,4,5-trisphosphate (PtdIns(3,4,5)P3) to produce PtdIns(3,4)P2, to negatively regulate PI3K (phosphoinositide 3-kinase) pathways. It acts as a negative regulator of B-cell antigen receptor signalling, and mediates signalling from the FC-gamma-RIIB receptor (FCGR2B) as well as a negative regulator of myeloid cell proliferation/survival and chemotaxis, neutrophil migration, mast cell degranulation, immune cells homeostasis, integrin alpha-IIb/beta-3 signalling in platelets and JNK signalling in B-cells. It regulates proliferation of osteoclast precursors, macrophage programming, phagocytosis and activation and is required for endotoxin tolerance. It may also hydrolyze PtdIns(1,3,4,5)P4. It interacts with adaptor proteins such as SHC, GABs, and DOKs. It features an N-terminal SH2 domain, a central catalytic 5-phosphatase domain, and a C-terminal proline-rich region. SHIP1 is primarily expressed in immune cells like B cells, mast cells, and myeloid cells. Primarily cytoplasmic but recruited to the plasma membrane upon receptor activation. It is expressed in microglial cells, where it regulates immune response, making it a key risk factor and therapeutic target for Alzheimer's disease.
