Target Full Name: Dual specificity protein phosphatase 9

Target Alias: DUS9; EC 3.1.3.16; Mitogen-activated protein kinase phosphatase 4; MKP4

Product Type Specific: DUSP9 protein phosphatase pan-specific antibody

Antibody Code: NP043-2

Antibody Target Type: Pan-specific

Protein UniProt: Q99956

Protein SigNET: DUSP9

Antibody Type: Polyclonal

Antibody Host Species: Rabbit

Antibody Immunogen Source: Human DUSP9 (MKP4) sequence peptide Cat. No.: PE-01BBK85

Antibody Immunogen Sequence: RLRRGSLSVRALLPGC

Antibody Immunogen Description: Corresponds to amino acid residues R53 to G67.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Antibody Modification: Unconjugated. Contact KInexus if you are interest in having the antibody biotinylated or coupled with fluorescent dyes.

Antibody Concentration: 1 mg/ml

Storage Buffer: Phosphate buffered saline pH 7.4, 0.05% Thimerasol

Storage Conditions: For long term storage, keep frozen at -40°C or lower. Stock solution can be kept at +4°C for more than 3 months. Avoid repeated freeze-thaw cycles.

Product Use: Western blotting | Antibody microarray

Antibody Dilution Recommended: 2 µg/ml for immunoblotting

Antibody Species Reactivity: This antibody detects the target phosphoprotein in the following species due to conservation of amino acid sequence: Human | Chimpanzee | Dog.

Antibody Positive Control: The observed molecular mass of the processed target protein on SDS-PAGE gels is reported to be around 37-43 kDa.

Antibody Specificity: High

Scientific Background: DUSP9 (MKP4) is a dual specificity phosphatase that dephosphorylates MAP kinases on phospho-tyrosine and phospho- threonine residues and negatively regulates their phosphotransferase activity in response to changes in the cellular environment. It primarily targets the ERK family and to a lesser degree the p38 and JNK MAP kinases. It is critical for controlling MAPK pathway cascades, influencing processes like cell stemness, stress response, and metabolic regulation. It features a C-terminal catalytic domain and an N-terminal Cdc25-like (CH2) domain. The protein is found in both the cytoplasm and the nucleus. Its expression is high in certain tissues, such as the placenta, and it is implicated in placental development, with its absence leading to lethal defects in embryo development. Aberrant expression of DUSP9 is linked to diseases such as liver metabolic syndrome, obesity, diabetes, cardiac dysfunction, and cancer progression (e.g., in hepatocellular carcinoma). It can act as a regulator of tumor progression, with its downregulation often associated with increased tumor growth, while its overexpression can promote apoptosis in certain cancer types.
