Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: DUSP8 (hVH-5) is a dual specificity phosphatase that exhibits activity towards both tyrosine-protein phosphate as well as with serine/threonine-protein phosphate. It features a Rhodanese domain (kinase interaction motif) and a tyrosine-protein phosphatase domain (active site). It acts as a negative regulator of MAPK signaling pathways (JNK, p38, and ERK) by dephosphorylating their phosphotyrosine and phosphoserine/threonine residues. It is highly expressed in the brain (and especially developing brain), heart, and skeletal muscle, where it functions as a cytoplasmic "gatekeeper" involved in cellular stress, proliferation, apoptosis, and metabolic regulation. It acts as a negative regulator of stress-induced ERK1/2 signalling in cardiac myocytes and regulates hypothalamic JNK signaling, influencing insulin sensitivity. Its expression is induced by nerve growth factor and insulin. It regulates metabolic effects induced by JNK activation in obesity. DUSP8 is linked to type 2 diabetes risk and is involved in colorectal and breast cancer progression, as well as cardiac ventricular remodeling.
