Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: DUSP6 (MKP3) is a dual specificity phosphatase that dephosphorylates MAP kinase ERK1 (and ERK2) on both 'Thr-202' and 'Tyr-204', and inhibits its activity during mitosis following initial growth factor activation. DUSP6 plays complex roles in cellular differentiation, development, and as both a tumour suppressor or pro-oncogenic factor depending on cancer type. It features a C-terminal catalytic domain, an N-terminal Cdc25-like (CH2) domain, a kinase interaction motif (KIM) and a nuclear export signal (NES). Unlike many other dual-specificity phosphatases, DUSP6 is mainly localized in the cytoplasm. It is expressed at high levels in the heart and pancreas. Mutations in DUSP6 are associated with congenital hypogonadotropic hypogonadism, which can include symptoms like anosmia (loss of smell) and hearing loss. DUSP6 is implicated in Alzheimer's disease, where reduced levels are correlated with cognitive decline. It functions as a tumour suppressor in cancers like pancreatic and lung cancer, but can act in a pro-oncogenic manner in others, such as glioblastomas, thyroid carcinomas and breats cancer.
