Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: CDC25A (Cell Division Cycle 25A) is a 59 kDa protein-tyrosine phosphatase that functions as a dosage-dependent inducer of mitotic progression. It removes inhibitory phosphates from CDKs, promoting G1/S and G2/M phase transitions. It directly dephosphorylates CDK1 (at Thr-14 and Tyr-15) and stimulates its kinase activity. It also dephosphorylates CDK2 in complex with cyclin E. It contains 524 amino acids with a catalytic rhodanese homology domain at the C-terminus and a regulatory N-terminus. CDC25A is activated by PIM1, and inactivated through phosphorylation by checkpoint kinases (Chk1/Chk2). CDC25A is degraded via the ubiquitin-proteasome pathway (mediated by Chk1/Chk2) in response to DNA damage, stopping cell division, which allows for DNA repair and acts as a checkpoint. While often a survival factor, it can act as a pro-apoptotic protein when cleaved by caspases during stress. It has a highly conserved C-terminal catalytic domain and a variable N-terminal regulatory domain.
