Scientific Background: Proteins in this family participate in pathways regulated by reversible phosphorylation at serine and threonine residues, potentially playing a role in the regulation of cell growth and differentiation. PP5 (Protein Phosphatase 5 ; PPT) is a unique, widely expressed serine/threonine phosphatase that regulates critical cellular processes, including stress responses, DNA damage repair, and steroid hormone signaling. It is a 58 kDa protein featuring a tetratricopeptide repeat (TPR) domain that enables it to act as a co-chaperone with Hsp90. PP5 is predominantly a nuclear protein and interacts with CDC16 and CDC27. It is found in association with several proteins that influence intracellular signlaing-cascades intitiated by hormones (glucocorticoids) or cellular stress (hypoxia, oxidative stress and DNA Damage). It phosphorylates serine residues of skeletal muscle phorylatse and histone H1. It may also be involved in mitosis and RNA biogenesis regulation (1). It regulates MAPK-mediated growth, cell cycle checkpoint activation, and apoptosis inhibition, specifically via p53 and ATM/ATR pathways. PP5 is heavily involved in cancer progression (notably breast and renal cell carcinoma) by regulating Hsp90-dependent kinases and providing a survival advantage to tumour cells.
