Scientific Background: PPP5C (Serine/threonine-protein phosphatase 5) is a protein phosphatase that dephosphorylates a myriad of proteins involved in different signaling pathways including the kinases CSNK1E, ASK1/MAP3K5, PRKDC and RAF1, the nuclear receptors NR3C1, PPARG, ESR1 and ESR2, SMAD proteins and TAU/MAPT. Proteins in this family participate in pathways regulated by reversible phosphorylation at serine and threonine residues, potentially playing a role in the regulation of cell growth and differentiation. PP5 or PPT is predominantly a nuclear protein and interacts with CDC16 and CDC27. It is found in association with several proteins that influence intracellular signlaing-cascades intitiated by hormones (glucocorticoids) or cellular stress (hypoxia, oxidative stress and DNA Damage). It phosphorylates serine residues of skeletal muscle phosphorylase and histone H1. It may also be involved in mitosis and RNA biogenesis regulation (1).
