Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: DUSP2 (PAC1) is a dual specificity phosphatase that dephosphorylates and inactivated MAP kinases (ERK, p38, JNK). For example, it dephosphorylates ERK2 on both 'Thr-183' and 'Tyr-185', and inhibits its activity during mitotic cell cycle progression following initial growth factor activation. It limits inflammation by regulating STAT3 signalling and TH17 cell differentiation. It is primarily localized to the nucleus of cells in hematopoietic tissues, where it controls immune responses. Its expression can be induced by p53 to promote apoptosis. It acts as a tumour suppressor, and its reduced expression is linked to poor prognosis in various cancers (e.g., cervical, colon). DUSP2 deficiency can lead to prolonged MAPK signalling, which can promote cancer stem cell accumulation and chemoresistance, and decreased immune-mediated inflammation.
