Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: CDC25C (Cell Division Cycle 25C) is a protein-tyrosine phosphatase that functions as a dosage-dependent inducer of mitotic progression. When phosphorylated, it is highly effective in activating G2 cells into prophase. It directly dephosphorylates CDK1 and CDK2 (at Thr-14 and Tyr-15, which are phosphorylated by checkpoint kinases like CHK1 and Wee1 kinase) and activates their kinase activities. It also acts as a DNA damage checkpoint enzyme to ensures proper DNA repair before mitosis. Its phosphatase activity is heavily controlled through phosphorylation and dephosphorylation, specifically through kinases like PLK1/Aurora A and check-point kinases ATR/CHK1. While active in the nucleus during mitosis, it is also found in the cytoplasm and associated with centrosomes.
