Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: CDC25B (Cell Division Cycle 25B) is a protein-tyrosine phosphatase that functions as a dosage-dependent inducer of mitotic progression. It is required for G2/M phases of the cell cycle progression and abscission during cytokinesis in a ECT2-dependent manner. It directly dephosphorylates CDK1 (Thr-14 and Tyr-15) and stimulates its kinase activity. Its expression increase in G2 phase and peaks in metaphase, with degradation occurring upon mitotic exit. It shuttles between the nucleus (M and G1 phases) and the cytoplasm (S and G2 phases). It is regulated by phosphorylation (e.g., by MAPKAP kinase-2 and p38) and 14-3-3 protein binding, which inhibits its phosphatase activity. It has three 14-3-3 binding sites (Ser-323, Ser-230, and Ser-151).
