Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: PCYT1B (Phosphate cytidylyltransferase 1B, choline) is the rate limiting enzyme in the biosynthetic pathway for phosphatidylcholine. It catalyzes the reaction: CTP + phosphocholine = diphosphate + CDP-choline. It is an ambiquitous enzyme that resides in the cytoplasm of cells in an inactive form. Diacylglycerol, acidic phospholipids and fatty acids drive the binding of PCYT1B to the endoplasmic reticulum and stimulation of its catalytic activity. It is critical for membrane synthesis and lipid metabolism, with high expression in the brain, placenta, ocary, testis, and fetus. It belongs to the cytidylyltransferase family and possesses a catalytic core and helical domain nearly identical to PCYT1A, but lacks the specific nuclear localization signal found in PCYT1A. Multiple alternative splicing variants exist. The protein can be expressed in 35-kD and 42-kD forms, with 42-kD potentially being a post-translationally modified version. It is often found in the cytoplasm and associates with the endoplasmic reticulum. It plays a role in phosphatidylcholine compositional homeostasis, and has been associated with studies on neonatal respiratory distress syndrome and preeclampsia.
