Antibody Immunogen Description: Corresponds to amino acid residues K528 to D543. Neurofilaments usually contain three intermediate filament proteins: L, M, and H which are involved in the maintenance of neuronal caliber.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: NF-L (NEFL; neurofilament light chain) is a 61-68 kDa cytoskeletal protein that belongs to the intermediate filament family and a structural protein in axons. Neurofilaments usually contain three intermediate filament proteins: L, M, and H which are involved in the maintenance of neuronal caliber. NF-L is the most abundant of the three. It is essential for maintaining axonal caliber, shape, and stability. It likely cooperates with the neuronal intermediate filament proteins PRPH and INA to form neuronal filamentous networks, which are essential for axon development, neurotransmission, and radial axon diameter. It features a tripartite structure (a head, a central alpha-helical rod domain, and a tail) that allows it to polymerize and provide structural integrity. It is found in the cytoplasm, axon, and neuron projections.
