Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: CPD (Carboxypeptidase D) is an integral 180 kDa membrane metallocarboxypeptidase that belongs to the peptidase M14 family. It features three tandem carboxypeptidase domains and a transmembrane region, with optimal activity at pH 5–7., and requires zinc ions. It functions in the processing of proteins and peptides in the secretory pathway. Mechanistically, it cleaves exclusively C-terminal basic residues, including Arg and Lys. By removing terminal residues, it can modulate the activity, stability, and receptor-binding properties of bioactive peptides. It is highly expressed in eukaryotic secretory pathways, particularly the trans-Golgi network (TGN), and is crucial for maturation of peptide hormones. It is highly expressed in placenta, pancreas and hepatoma cells, with lower levels found in skeletal muscle, heart and colon carcinoma and melanoma cell lines. CPD possesses three tandem carboxypeptidase domains (two active, one inactive) and is a homologue of the hepatitis B virus-binding protein. It is exploited as an alternative entry receptor for adeno-associated virus (AAV). CPD expression is influenced by hormones like testosterone and estrogen, as well as growth factors.
