Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: 14-3-3 sigma is an adapter protein implicated in the regulation of a large spectrum of both general and specialized signalling pathways. It binds to a large number of partners through recognition of specific phosphoserine or phosphothreonine motifs. Its binding generally results in the modulation of the activity of the binding phosphoprotein partner. It promotes cytosolic retention of GBP1 GTPase by binding to phosphorylated GBP1, thereby inhibiting the innate immune response. It also acts as a TP53/p53-regulated inhibitor of G2/M progression. When bound to KRT17, 14-3-3 sigma regulates protein synthesis and epithelial cell growth by stimulating Akt/mTOR pathway. It acts to maintain desmosome cell junction adhesion in epithelial cells via interacting with and sequestering PKP3 to the cytoplasm, thereby restricting its translocation to existing desmosome structures to maintain desmosome protein homeostasis. It also acts to facilitate PKP3 exchange at desmosome plaques to maintain keratinocyte intercellular adhesion.
