Scientific Background: IFT88 (Intraflagellar transport protein 88 homolog; polaris) is a 93 kDa protein in the IFT Complex B essential for cilia assembly, maintenance, and signalling (e.g., in the Hedgehog signaling pathway). It appear to be involved in primary cilium biogenesis, and is also implicated in autophagy, since it is required for trafficking of ATG16L and the expansion of the autophagic compartment. It is a tetratricopeptide (typically 12) repeat (TPR) protein, through which it is likely involved in multiple protein-protein interactions. It interacts with other IFT complex B proteins, such as IFT20, IFT74, and IFT81. Beyond cilia, IFT88 has roles in cell-cycle regulation (G1-S transition), mitosis, and mediating cell polarization during migration. It is expressed in the heart, brain, liver, lung, kidney, skeletal muscle and pancreas. It is located in the cytoplasm, centrosomes, cilia basal bodies, and along the cilia body. Mutations in this region are linked with defective cilia, ciliopathies, polycystic kidney disease (PKD), retinal degeneration, and left-right asymmetry.
