Scientific Background: Autophagy related 4A, cysteine peptidase (ATG4A or APG4A) is a C-54 cysteine protease that also belongs to the family of autophagin proteins, and is primarily found in skeletal muscle and brain. ATG4A mediates cell remodeling and homeostasis, and is often involved in cancer development and cellular stress responses. It has a high preference for GABARAPL2, followed by MAP1LC3A and GABARAP. ATG4A primes pro-LC3 and GABARAP proteins by cleaving their C-termini to expose a glycine residue necessary for their membrane insertion It cleaves ATG8 family proteins to enable their conjugation to phosphatidylethanolamine, a crucial step for autophagosome formation. It cleaves GATE-16. Mutations in the ATG4a gene is critical for redox regulation and inhibits autophagosome formation. ATG4A is important for maintaining a stem-like phenotype in breast cancer cells. It has also been linked to promoting tumour metastasis in other contexts. TG4A may be a potential target in cataract therapies in view of its role in lens degradation.
