Scientific Background: Beclin 2 is a 49 kDa novel coiled-coil protein related to the autophagic Beclin 1 protein. It features a BH3 domain, a coiled-coil domain, and a C-terminal domain. It functions in two distinct lysosomal degradation pathways: autophagy and the degradation of G protein-coupled receptors (GPCRs) via interaction with GASP1. It is thought to interact with Bcl-2, an anti-apoptotic protein. It has also been found to mediate G protein-coupled receptor (GPCR) degradation (1). While both Beclin 1 and Beclin 2 regulate autophagy, Beclin 2 uniquely facilitates the agonist-induced lysosomal degradation of specific GPCRs (such as CB1R and DOR). Becilin 2 acts as a negative regulator of innate immune signalling, and mediates the degradation of inflammasome sensors such as AIM2 and NLRP3. It is predominantly expressed in the brain, heart, skeletal muscle, placenta, and uterus, with distinct expression patterns from Beclin 1. Beclin 2 is involved in metabolism, as haploinsufficiency (loss of one allele) in mice results in increased food intake, obesity, and insulin resistance, partly due to reduced degradation of the cannabinoid receptor 1 (CB1R).
