Scientific Background: The alpha 2C Adrenergic Receptor (a2C-AR) is a G protein-coupled receptor (GPCR) that mediates sympathetic nervous system activity. It functions primarily as a presynaptic autoreceptor, regulating neurotransmitter release at low nerve activity levels. It is predominantly found in the central nervous system, particularly the basal ganglia, and plays a role in cold-induced vasoconstriction. a2C-AR controls the release of neurotransmitter from central adrenergic neurons and from sympathetic nerves in the heart. It inhibits norepinephrine release in the central nervous system and sympathetic nerves. This receptor also plays a role in cognitive and behavioral function. It acts as an inhibitory receptor, coupled with Gi/o proteins to inhibit adenylyl cyclase, which suppresses cAMP production. It promotes ERK1/2 stimulation and suppresses voltage-gated calcium channels. It features 7-transmembrane helical bundle (TM I–VII), three extracellular loops (ECL1–3), three intracellular loops (ICL1–3), and a C-terminal helix VIII. Two variants are produced by alternative splicing. a2C-AR has been reported extensively in the brain. Blood, corpus cavermosum, kidney, and heart tissues have also been reported to express the protein. It affects the regulation of sympathetic adrenergic stimulation and plays a role in the pathogenesis of conditions like Raynaud's phenomenon. It is implicated in psychiatric conditions, such as schizophrenia or bipolar disorder. This description may include information annotated by UniProt and/or Google AI.
