Scientific Background: VGLUT3 (Vesicular Glutamate Transporter 3) is a 60-62 kDa multipass membrane protein restricted to synaptic vesicles of glutamergic neurons. It is specifically expressed in amygdala, cerebellum, hippocampus, medulla, spinal cord, and thalamus. It functions as a proton-driven pump (uniporter) using the electrochemical gradient created by v-ATPase to fill vesicles with glutamate. It is responsible for packing the excitatory neurotransmitter glutamate into synaptic vesicles. Unlike its counterparts VGLUT1 and VGLUT2, which are found in traditional excitatory neurons, VGLUT3 is considered an "atypical" transporter, largely expressed in cells that typically utilize other neurotransmitters (e.g., serotonin, acetylcholine, or GABA). VGLUT3 plays a critical role in modulating behavior, specifically in anxiety, depression, social interaction, and motor control. Human VGLUT3 shares a 72% sequence homology with VLGUT2 and BNPI. Impairment or mutation in the SLC17A8 gene is linked to non-syndromic deafness DFNA25. Mice lacking VGLUT3 are viable but exhibit increased anxiety-associated behaviors and, in some models, improved motor function in Parkinson's models.
