Antibody Dilution Recommended: WB (1:1000); optimal dilutions for assays should be determined by the user.

Antibody Potency: Detects a ~97 kDa protein, non-glycosylated version at 68 kDa protein in cell and tissue lysates by Western blotting. Other minor bands associated with hNIS at 160 kDa protein, and degradation products at ~30 kDa protein, and ~15 kDa protein in cell and tissue lysates by Western blotting.

Antibody Species Reactivity: This antibody detects the target protein in the following species due to conservation of amino acid sequence: Human | Rat | Mouse.

Antibody Positive Control: 1 µg/ml of SMC-390 was sufficient for detection of hNIS in 20 µg of transfected COS-7 cell membrane lysate by ECL immunoblot analysis using Goat anti-mouse IgG:HRP as the secondary antibody.

Scientific Background: The sodium iodide symporter (NIS), encoded by the SLC5A5 gene, is an integral membrane glycoprotein (approx. 87 kDa) that actively transports iodide from the bloodstream across the basolateral membrane into thyroid epithelial follicular cells (1, 2). This is important step in the process of iodide organificaton and the formation of triiodothyronine and thyroxine, making NIS crucial for regulating body metabolism and growth (3). It is a transmembrane protein containing 13 transmembrane domains, an extracellular N-terminus, and an intracellular C-terminus. It typically forms dimers stabilized by disulfide bridges. NIS acts as a symporter, utilizing the sodium gradient (maintained by the Na+/K+ATPase pump) to move two sodium cations Na+ alongside one iodide anion (I-)into the cell. While found primarily on the basolateral membrane of thyroid cells, NIS is also expressed during lactation in breast tissue, as well as in the salivary glands, ovaries, and gastric mucosa.
