Scientific Background: The JNK-interacting protein (JIP) group of scaffold proteins selectively mediates JNK signalling by aggregating specific components of the MAPK cascade to form a functional JNK signalling module. JIP2 binds to components of the JNK pathway (JNK1/2/3, MKK7), as well as motor proteins (kinesin light chain, KLC1) for transportation. It inhibits IL1 beta-induced apoptosis in insulin-secreting cells. JIP2 may function as a regulator of vesicle transport, through interactions with the JNK-signalling components and motor proteins. JIP2 features long N-terminal regions with interaction sites for kinases and a C-terminal domain (SH3 and PID domains) for scaffolding, sharing similar structure with JIP1. It is expressed mainly in the cerebellum, pituitary gland, occipital lobe, and the amygdala of the brain, but also in the pancreas, including insulin-secreting cells. JIP2 is linked to Phelan-McDermid syndrome (due to its role in regulating NMDA/AMPA receptor ratios) and is involved in glucose homeostasis.
