Scientific Background: GABRB1 is a beta subunit of the heteropentameric ligand-gated chloride channel gated by gamma-aminobutyric acid (GABA). GABA-gated chloride channels (GABA(A) receptors (GABAAR)), consist of five subunits arranged around a central pore and contain one or two GABA active binding sites located at the alpha and beta subunit interfaces, depending on subunit composition. The individual subunits of these receptors have similar sequences and structural features (1). GABA-A receptors are the major fast inhibitory neurotransmitter gated ion channels in the brain (2). When activated by GABA, GABAARs selectively allow the flow of chloride anions across the cell membrane. Beta-containing GABAARs can simultaneously bind GABA and histamine where histamine binds at the interface of two neighboring beta subunits, which has been implicatedthe regulation of sleep and wakefulness. It is found in the plasma membrane, dendrites, and synapse of neurons. Beta-1 containing GABAA receptors are found in regions controlling reward-related behavior, and their dysfunction can contribute to addictive behaviors. Mutations in GABRB1 are linked to Developmental and Epileptic Encephalopathy 45 (DEE45), a condition characterized by severe early-childhood epilepsy and cognitive impairment. GABRB1 expression is altered in the cerebellum of individuals with schizophrenia, bipolar disorder, and major depression, potentially affecting GABAergic signalling.
