Scientific Background: GABRA5 is an alpha subunit of the heteropentameric ligand-gated chloride channel gated by gamma-aminobutyric acid (GABA). The GABA-A receptor is a member of the superfamily of fast acting ligand-gated ion channels. The individual subunits of these receptors have similar sequences and structural features (1). GABA-A receptors are the major fast inhibitory neurotransmitter gated ion channels in the brain (2). GABAARs containing alpha-5/GABRA5 subunits are mainly extrasynaptic and contribute to the tonic GABAergic inhibition in the hippocampus. Extrasynaptic alpha-5-containing GABAARs in CA1 pyramidal neurons have been implicated in learning and memory processes. Inverse agonists of the Alpha5 subunit may be useful in the treatment of Alzheimer's Disease (3). Mutations in GABRA5 are linked to early-onset epilepsy and developmental delays.
