Scientific Background: GABRA2 is an alpha subunit of the heteropentameric ligand-gated chloride channel gated by gamma-aminobutyric acid (GABA). The GABA-A receptor is a member of the superfamily of fast acting ligand-gated ion channels. When GABA binds, the receptor opens, allowing chloride ion influx, which inhibits neuron activity and reduces nervous transmission. The individual subunits of these receptors have similar sequences and structural features (1). GABA-A receptors are the major fast inhibitory neurotransmitter gated ion channels in the brain (2). The alpha-2 subunit exhibits synaptogenic activity together with beta-2 and very little to no activity together with beta-3. The gamma-2 is subunit necessary but not sufficient to induce rapid synaptic contacts formation. When GABA binds, the receptor opens, allowing chloride ion influx, which inhibits neuron activity and reduces nervous transmission.
