Scientific Background: Chapsyn-110 (PSD93, DLG2) is a part of the membrane-associated putative guanylate kinase (MAGUK) family. This 110 kDa neuroal scaffolding protein binds directly to the N-methyl-D-aspartate (NMDA) receptor, and Shaker K+ channel subunits (1). It associated tightly with the postsynaptic density in the brain, often in a somatodendritic pattern, and mediates the clustering of both NMDA receptors and K+ channels in heterologous cells. The encoded protein forms a heterodimer with PSD-95 that may interact at postsynaptic sites to form a multimeric scaffold for the clustering of receptors, ion channels, and associated signalling proteins (1, 2). It has a similar domain organization to PSD-95/SAP90 (70–80% similarity), containing three PDZ domains, an SH3 domain, and a guanylate kinase domain. Deficiency in tChapsyn-110 can lead to reduced synaptic long-term potentiation (LTP) in the hippocampus.
