Scientific Background: Beclin 1 is a ubiquitous, peripherlal membrane-bound, adaptor/scaffold protein that belongs to the beclin family. It plays a central role in autophagy and endocytosis. It functions as core subunit of the PI3K complex that mediates formation of phosphatidylinositol 3-phosphate. Different complex forms are thought to play a role in multiple membrane trafficking pathways. It is important for initiating autophagy (cellular self-eating), a process for degrading damaged organelles to maintain cellular homeostasis. It also regulates apoptosis (programmed cell death). PI3KC3-C1 is involved in initiation of autophagosomes and PI3KC3-C2 in maturation of autophagosomes and endocytosis. It mediates regulation of degradative endocytic trafficking and is required for the abcission step in cytokinesis, probably in the context of PI3KC3-C2. It is essential for the formation of PI3KC3-C2 but not PI3KC3-C1 PI3K complex forms.
