Scientific Background: ATG13 (Autophagy-related protein 13) is an autophagy factor that belongs to the ATG13 family, Metazoan subfamily. It is required for autophagosome formation and mitophagy. It is a target of the mTOR kinase signaling pathway that regulates autophagy through the control of the phosphorylation status of ATG13 and ULK1, and the regulation of the ATG13-ULK1-RB1CC1 complex. ATG13 is a heavily phosphorylated protein that acts as an adapter and scaffolding agent in the initiation of autophagy and selective mitophagy. It forms a core complex with ULK1, RB1CC1/FIP200, and ATG101, acting as a direct target of the TOR kinase signaling pathway to regulate autophagosome assembly. Under nutrient-rich conditions, mTORC1 phosphorylates ATG13, inhibiting its activity. Upon starvation, this inhibition is lifted, allowing ATG13 to facilitate autophagosome formation. Through its regulation of ULK1 phosphotransferase activity, ATG13 also plays a role in the control of the activity of mTORC1 and cell proliferation. It features an N-terminal HORMA domain (residues 1–268) and a C-terminal intrinsically disordered region (IDR) (residues 269–738). Its HORMA domain mediates the recruitment of downstream proteins, including ATG14 and Atg8 family proteins. The IDR domain mediates interactions with the TOR signaling complex. Mutations and deficiencies in ATG13 lead to impaired autophagic flux. It is also linked to non-autophagic processes, such as cardiac development.
