Scientific Background: ATG5 (Autophagy-related protein 5) is an E3-like ligase in the autophagy pathway, and essential for autophagosome formation and elongation. It is important for cellular homeostasis, quality control, pathogen resistance, and immune regulation. It forms a complex with ATG12 and ATG16L1 to facilitate the conjugation of LC3-I to PE to form LC3-II. ATG5 is involved in autophagic vesicle formation. It conjugates with ATG12 in a ubiquitin-like conjugating system that involves ATG7 as an E1-like activating enzyme and ATG10 as an E2-like conjugating enzyme. The ATG12-ATG5 conjugate acts as an E3-like enzyme, which is required for lipidation of ATG8 family proteins and their association to the vesicle membranes. ATG5 is involved in mitochondrial quality control after oxidative damage, and in subsequent cellular longevity.
