Scientific Background: ATF4 (Cyclic AMP-dependent transcription factor ATF-4) is a basic leucine-zipper (bZip) transcription factor, which regulates amino acid metabolism, DNA damage repair, chromatin remodeling, and apoptosis in response to cellular and ER stress. ATF4 works with various proteins, such as C/EBP homology protein (CHOP), aspargine synthetase (ASNS), and cAMP response element (CRE) among others to mediate cellular stress. ATF4 also regulates glucose homeostasis by suppressing beta-cell proliferation and insulin production. Furthermore, ATF4 targets the histone demethylase JMJD3 to alter chromatin structure and enhance gene transcription in response to amino acid deprivation. Under stress, its translation is preferential following the inactivation of initiation factor eIF2, allowing it to increase in concentration and regulate gene expression, often forming dimers with CHOP or CREB1. It features an bZIP domain for DNA binding/heterodimerization, a leucine zipper II (LZII) domain, and a PCAF domain. ATF4 acts as a pro-survival factor in specialized cells (e.g., osteoblasts, plasma cells) but can be pro-apoptotic (e.g., in neurons). It is involved in neurodegenerative diseases, tumour survival, and metabolic disorders (e.g., diabetes).Its activation is often found in cancer, promoting cell survival in hypoxic or nutrient-deprived tumors.
