Target Full Name: Pyruvate dehydrogenase kinase isoform 2; [Pyruvate dehydrogenase [lipoamide]] kinase isozyme 2, mitochondrial

Target Alias: Kinase Pyruvate dehydrogenase kinase 2; PDK2; Pyruvate dehydrogenase [lipoamide] kinase isozyme 2, mitochondrial precursor

Product Type Specific: PDHK2 (PDK2) protein kinase pan-specific antibody

Antibody Code: NN180-1

Antibody Target Type: Pan-specific

Protein UniProt: Q15119

Protein SigNET: PDK2

Antibody Type: Polyclonal

Antibody Host Species: Rabbit

Antibody Immunogen Source: Human PDK2 (PDHK2) sequence peptide Cat. No.: PE-01AZZ90

Antibody Immunogen Sequence: MRWVWALLKNASLAC

Antibody Immunogen Description: Corresponds to amino acid residues M1 to A14; N-terminus.

Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Antibody Modification: Unconjugated. Contact KInexus if you are interest in having the antibody biotinylated or coupled with fluorescent dyes.

Antibody Concentration: 1 mg/ml

Storage Buffer: Phosphate buffered saline pH 7.4, 0.05% Thimerasol

Storage Conditions: For long term storage, keep frozen at -40°C or lower. Stock solution can be kept at +4°C for more than 3 months. Avoid repeated freeze-thaw cycles.

Product Use: Western blotting | Antibody microarray

Antibody Dilution Recommended: 1 µg/ml for immunoblotting

Antibody Potency: Medium-weak immunoreactivity with recombinant human PDK2 on protein dot blots.

Antibody Species Reactivity: This antibody detects the target protein in the following species due to conservation of amino acid sequence: Human | Chimpanzee | Rat | Mouse.

Antibody Positive Control: The observed molecular mass of the processed target protein on SDS-PAGE gels is reported to be around 42-50 kDa.

Antibody Specificity: High

Antibody Cross Reactivity: No immunoreactivity on protein dot blots with recombinant human PDK1, PDK3 and PDK4.

Scientific Background: PDHK2 (PDK2, pyruvate dehydrogenase kinase 2) is a 72-kDa mitochondrial protein-serine/threonine kinase of the Atypical group and PDHK family. It is an essential kinase in the regulation of fatty acid and glucose metabolism by phosphorylating and inactivating the enzyme pyruvate dehydrogenase (PDC). It phosphorylates the E1 component of the PDC, primarily at Ser-293, Ser-300, and Ser-232, leading to a down-regulation of its activity. It acts as a gatekeeper in the mitochondria, controlling the rate of carbohydrate oxidation and directing pyruvate away from the TCA cycle, which is essential for maintaining carbohydrate homeostasis. PDHK2 activity is regulated by metabolic factors like pyruvate, NAD+, NADH, CoA, and acetyl-CoA. It can be inhibited by low levels of hydrogen peroxide (H2O2), acting as a sensor for oxidative stress and potentially increasing PDC activity during high mitochondrial respiration. PDHK2 features specialized binding sites, including a unique pyruvate binding site and a site for the inner lipoyl-bearing domain (L2) of the dihydrolipoyl transacetylase (E2) component of the PDC. PDHK2 expression is implicated in diseases such as diabetes and cancer. Overexpression of PDHK2 contributes to the Warburg effect (increased glycolysis) in cancer cells.
