Scientific Background: ULK3 (UNC-51 like kinase 3) is a ~58 kDa serine/threonine-protein kinase that regulates critical cellular processes, including Sonic hedgehog (SHH) signaling, autophagy, and cytokinetic abscission. It acts as a molecular switch, activating GLI transcription factors (GLI1, GLI2, GLI3) in the SHH pathway by promoting their nuclear translocation, and, when stressed, phosphorylates ESCRT-III complex components to control cell division timing. ULK3 controls the timing of cytokinetic abscission (the final separation of daughter cells) by binding to IST1 and phosphorylating ESCRT-III, particularly during cellular stress, such as lagging chromosomes or nuclear pore defects. It is involved in the regulation of autophagy, the cellular recycling process, particularly in response to senescence. It features an N-terminal kinase domain and a C-terminal regulatory domain. Its catalytic activity is regulated by phosphorylation in the kinase domain. Increased expression is associated with cancer-associated fibroblasts (CAFs) and tumor-enhancing properties, particularly in squamous cell carcinomas.
