Scientific Background: ULK2 (UNC-51 like kinase 2) is a serine/threonine protein kinasr that acts as a key regulator of autophagy in response to nutrient starvation, promoting cellular homeostasis. It works early in the autophagy pathway to initiate autophagophore formation and also plays a critical role in neuronal development, particularly in axon formation. It contains an amino-terminal kinase domain followed by a central proline-serine rich domain and a highly conserved carboxy-terminal domain. ULK2 serves as a downstream effector of AMPK and a regulator of mTORC1, often functioning alongside ULK1 in initiating autophagy. It also acts as a negative regulator of mTORC1 and AMPK signaling pathways. It has been linked to axon growth and is essential for autophagy. Structurally, ULK2 is similar to ATG1. It is highly expressed in the brain, playing a crucial role in endocytosis, axonal elongation, and neuronal differentiation.
