Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: Tau tubulin kinase 2 (TTBK2) is a ubiquitously expressed 137 kDa protein serine-threonine kinase that belongs to the casein kinase (CK1) group. It is crucial for initializing ciliogenesis (primary cilia assembly) and maintaining cilia stability by regulating microtubule dynamics. TTBK2 acts as a critical regulator of ciliogenesis by initiating the assembly of the ciliary axoneme and, in some contexts, maintaining it. TTBK2 associates with microtubules and tracks the plus-ends, influencing microtubule depolymerizing activity. It controls the initiation of ciliogenesis by binding to the distal end of the basal body and promoting the removal of CCP110, which caps the mother centriole, leading to the recruitment of IFT proteins, which build the ciliary axoneme. It phosphorylates tau on Serine residues, beta-tubulin, and proteins associated with cilia, such as CEP164. It has some substrate preference for proteins that are already phosphorylated on a Tyr residue at the +2 position relative to the phosphorylation site. It is expressed in many tissues, with highest levels in the brain and testis.
