Scientific Background: CaMK2a (CaMKII) is a protein-serine/threonine kinase of the CaMK group and CaMK2 family. It is a calcium/calmodulin-activated protein kinase involved in neurotransmitter release and long term potentiation in neurons. It is highly abundant in the brain, particularly at excitatory synapses in the postsynaptic density. It acts as a key mediator of synaptic plasticity, learning, and memory by regulating long-term potentiation (LTP) and activating upon increased calcium levels. Autophosphorylation of Thr-286 allows the kinase to switch from a calmodulin-dependent to a calmodulin-independent state. It is also involved in regulation of cell cycle progression, T-cell activation, and cardiac function. CaMK2a resides in large multi-oligomeric protein complexes (12 subunits) with other CaMK2 subunits (beta, gamma and delta). Mutations in the CAMK2A gene are linked to neurodevelopmental disorders, specifically autosomal dominant and recessive intellectual disabilities.
