Antibody Immunogen Source: Human HGK (ZC1) sequence peptide Cat. No.: PE-01BFM
Antibody Immunogen Sequence: ANDSPAKSLVDIDLSSLRDC
Antibody Immunogen Description: Corresponds to amino acid residues A2 to D20; N-terminus.
Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.
Antibody Modification: Unconjugated. Contact KInexus if you are interest in having the antibody biotinylated or coupled with fluorescent dyes.
Antibody Concentration: 1 mg/ml
Storage Buffer: Phosphate buffered saline pH 7.4, 0.05% Thimerasol
Storage Conditions: For long term storage, keep frozen at -40°C or lower. Stock solution can be kept at +4°C for more than 3 months. Avoid repeated freeze-thaw cycles.

Scientific Background: ZC1 (Hepatocyte progenitor kinase-like/Germinal center kinase-like Kinase; HGK; MAP4K4) is a protein-serine/threonine kinase of the STE group and STE20 family. It may play a role in the response to environmental stress and cytokines such as TNF-alpha, and appears to act upstream of the JUN N-terminal pathway. ZC1 specifically activates the MAPK8/JNK signaling pathway, frequently functioning through the TAK1-MKK4/MKK7 kinase cascade. it is involved in regulating actin cytoskeletal organization, inhibiting adipogenesis (fat cell formation), and mediating TNF-alpha-induced insulin resistance. It features an N-terminal kinase domain, a coiled-coil domain, and a C-terminal hydrophobic leucine-rich citron homology (CNH) domain, which is crucial for protein-protein interactions. ZC1 can phosphorylate SMAD1 at Thr-322. Whole-body knockouts of the ZC1 gene in mice are embryonic lethal, indicating its vital role in development, specifically in mesodermal and somite formation. Insertional mutagenesis studies in mice support a role for this protein kinase in mouse cancer oncogenesis. ZC1 is widely expressed in tumour cell lines and plays a significant role in promoting cell transformation and invasion. It acts as a pro-migratory kinase often associated with poor prognosis in cancers such as pancreatic, colorectal, and lung adenocarcinoma.
