Scientific Background: ULK1 (UNC-51 like kinase 1) is a ~112-kDa serine/threonine protein kinase that acts as the master initiator of autophagy in mammals, serving as the orthologue to yeast Atg1. It forms a core complex with ATG13, FIP200, and ATG101 to trigger autophagosome formation in response to nutrient starvation, regulated by mTORC1 (inhibition) and AMPK (activation). ULK1 phosphorylates key components like BECN1 and VPS34 to initiate autophagy. It is crucial for selective autophagy, including mitophagy (mitochondrial clearance). When nutrients are plentiful, mTORC1 phosphorylates and inactivates ULK1. During starvation/energy stress, AMPK phosphorylates and activates ULK1.
