Production Method: The immunizing peptide was produced by solid phase synthesis on a multipep peptide synthesizer and purified by reverse-phase hplc chromatography. Purity was assessed by analytical hplc and the amino acid sequence confirmed by mass spectrometry analysis. This peptide was coupled to KLH prior to immunization into rabbits. New Zealand White rabbits were subcutaneously injected with KLH-coupled immunizing peptide every 4 weeks for 4 months. The sera from these animals was applied onto an agarose column to which the immunogen peptide was thio-linked. Antibody was eluted from the column with 0.1 M glycine, pH 2.5. Subsequently, the antibody solution was neutralized to pH 7.0 with saturated Tris.

Scientific Background: PKG2 (PRKG2) is an ~87 kDa protein-serine/threonine kinase of the AGC group and PKG family. It acts as a membrane-bound, cGMP-dependent receptor to mediate critical cellular signalling, particularly in intestinal water/electrolyte transport, renin release from kidney juxtaglomerular cells and endochondral ossification (bone growth). Its deficiency is associated with dwarfism and skeletal dysplasia. PRKG2 phosphorylates target proteins (e.g., CFTR, Sox9) in response to increased intracellular cGMP levels. It also plays a role in synaptic plasticity by acting downstream of NMDAR. PRKG2 is highly expressed in cells like chondrocytes, intestinal mucosa, and certain brain neurons. Variants of the PRKG2 gene are linked to Acromesomelic Dysplasia 4, Spondylometaphyseal Dysplasia (Pagnamenta Type), and general skeletal defects.
